Learn the fundamentals of CRISPR-Cas9 and make precision gene edits hands-on Join us at Counter Culture Labs for a 2-day bootcamp where you will learn the fundamentals of CRISPR-Cas9 and make precision gene edits in E. coli. CRISPR-Cas9 is a precision genetic engineering tool where guide RNA directs a Cas9 enzyme to a specific target to make a double-stranded break which can be repaired with template DNA. Not only will we genetically engineer microbes with CRISPR but also design our own edits and verify transformations with molecular biology techniques, including PCR and gel electrophoresis. Program Details: April 25 & 26 weekend from 10am - 4:30pm daily Day 1 History of CRISPR Plasmid DNA and inserting DNA into microbes Evolution of competence (techniques: heat shock, chemical, electroporation) Preparing electrocompetent cells and performing electroporation Selection vs screening and plating transformants Electronic lab notebook Day 2 Observing phenotypes Genotyping tools for verifying edits PCR to amplify transformant DNA Gel electrophoresis and sequencing Generating your own Cas9 sgRNA and template DNA design for inserting fluorescent proteins into microbes Advanced CRISPR applications and technologies Required lab attire for safety: Long pants and closed-toed shoes. ​ Please bring a laptop so that you can participate in the DNA design lessons. We will be hosting a screening of the Japanese-American drama "Hito Hata: Raise the Banner"(1980) on Sunday, April 26th as a fundraiser. Nostalgic For Nothing Cinema is excited to present the next Movie Night Fundraiser at Moments Co-op, featuring Hito Hata: Raise the Banner(1980, 93 min), directed by Robert A. Nakamura and Duane Kubo. A landmark project directed by Robert A. Nakamura and Duane Kubo, Hito Hata: Raise the Banner is the first feature-length film made by and about Asian Pacific Americans. Capturing the contributions and hardships of Japanese Americans from the turn of the twentieth century, the film centers on Oda (the late veteran actor/director Mako), a feisty issei (first-generation Japanese American) and elderly single laborer living in Los Angeles’s Little Tokyo. Through a series of flashbacks, the story traces Oda’s life, from his time as a laborer on the transcontinental railroad to his struggle to save the home of Little Tokyo’s residents when the community is threatened with redevelopment.
